3.1.1 Autofluorescence
The study of biological tissues using fluorescence microscopy faces several obstacles. All cells exhibit autofluorescence in the absence of an applied probe. The main band of emission in the yellow-green band, between 500 and 600 nm, is due to flavoproteins when excitation is in the blue around 450 to 480 nm. NADH emits in the blue (460 to 470 nm) when excited in the UV around 360 nm.
