4. Advanced microscopy techniques

In this section, we'll focus on microscopic measurements of fluorescence intensity, which constitute the techniques of fluorescence imaging. Measurements of other physical parameters of fluorescence, such as lifetime, dichroism, etc., will not be discussed.

4.1 Restricting the area explored

In this section, we will describe specific microscopy techniques for limiting the size of the "blur" zone created by a fluorescent source. The causes of blur are mainly due to the illumination of fluorescent objects outside the plane of focus and to the image spreading function of a point source ("PSF").
