It is difficult to define an exact methodology that will lead to the ideal solution, and this stage therefore requires several experiments. The device chosen must highlight the areas to be studied, in order to facilitate downstream processing and increase its efficiency and speed. It must be perfectly controlled and, in most cases, protected from ambient light.
