The use of a piston-operated volumetric apparatus (POVA) requires traceability to national or international standards. This paper seeks to enable a user to implement the necessary equipment in order to achieve metrological confirmation of a POVA. In this way, the POVA will be traceable and the process uncertainties will be controlled.

 INTRODUCTION

Using pipettes to transfer liquids is a daily activity in most life science research laboratories. From academic labs to state-of-the-art laboratories to testing labs applying standard operating procedures, the data generated can be strongly influenced by pipette performance and user technique.

A brief review of the scientific literature shows a constant stream of reminders that pipettes and pipetting technique play a major role in the success or failure of an experiment. Ignoring pipetting technique can result in a significant loss of time and money, crucial for any laboratory.

Modern quality assurance systems require periodic gravimetric control and, where necessary, pipette adjustment. To perform these checks efficiently and reliably, you need high-quality balances, software support and perfectly matched accessories. Together with pipettes and tips, these form an inseparable system, a kind of chain whose quality depends on that of the weakest link.

Pipette performance depends on a number of factors, including maintenance and periodic checks. Users must also develop their pipetting skills to ensure reliable data reproduction.

In addition to the quality of the pipette and tip, the operator's knowledge and skill are crucial. The aim of this article is to present the theoretical aspects of pipetting with air column pipettes.
